Two-stage stochastic programming with robust constraints for the logistics network post-disruption responsestrategy optimization

摘要: Logistics networks (LNs) are essential for the transportation and distribution of goods or services from suppliers to consumers. However, LNs with complex structures are more vulnerable to disruptions due to natural disasters and accidents. To address the LN post-disruption response strategy optimization problem, this study proposes a novel two-stage stochastic programming model with robust delivery time constraints. The proposed model jointly optimizes the new-line-opening and rerouting decisions in the face of uncertain transport demands and transportation times. To enhance the robustness of the response strategy obtained, the conditional value at risk (CVaR) criterion is utilized to reduce the operational risk, and robust constraints based on the scenario-based uncertainty sets are proposed to guarantee the delivery time requirement. An equivalent tractable mixed-integer linear programming reformulation is further derived by linearizing the CVaR objective function and dualizing the infinite number of robust constraints into finite ones. A case study based on the practical operations of the JD LN is conducted to validate the practical significance of the proposed model. A comparison with the rerouting strategy and two benchmark models demonstrates the superiority of the proposed model in terms of operational cost, delivery time, and loading rate.

Durability of neutralizing antibodies and T-cell response post SARS-CoV-2 infection

摘要: The ongoing pandemic of coronavirus disease 19 (COVID-19) is caused by a newly discovered β coronavirus named severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). How long the adaptive immunity triggered by SARS-CoV-2 can last is of critical clinical relevance in assessing the probability of second infection and efficacy of vaccination. Here we examined, using ELISA, the IgG antibodies in serum specimens collected from 17 COVID-19 patients at 6−7 months after diagnosis and the results were compared to those from cases investigated 2 weeks to 2 months post-infection. All samples were positive for IgGs against the S- and N-proteins of SARS-CoV-2. Notably, 14 samples available at 6−7 months post-infection all showed significant neutralizing activities in a pseudovirus assay, with no difference in blocking the cell-entry of the 614D and 614G variants of SARS-CoV-2. Furthermore, in 10 blood samples from cases at 6−7 months post-infection used for memory T-cell tests, we found that interferon γ-producing CD4 and CD8 cells were increased upon SARS-CoV-2 antigen stimulation. Together, these results indicate that durable anti-SARS-CoV-2 immunity is common in convalescent population, and vaccines developed from 614D variant may offer protection from the currently predominant 614D variant of SARS-CoV-2.

Modeling of the resilient supply chain system from a perspective of production design changes

摘要: Building an effective resilient supply chain system (RSCS) is critical and necessary to reduce the risk of supply chain disruptions in unexpected scenarios such as COVID-19 pandemic and trade wars. To overcome the impact of insufficient raw material supply on the supply chain in mass disruption scenarios, this study proposes a novel RSCS considering product design changes (PDC). An RSCS domain model is first developed from the perspective of PDC based on a general conceptual framework, i.e., function-context-behavior-principle-state-structure (FCBPSS), which can portray complex systems under unpredictable situations. Specifically, the interaction among the structure, state and behavior of the infrastructure system and substance system is captured, and then a quantitative analysis of the change impact process is presented to evaluate the resilience of both the product and supply chain. Next, a case study is conducted to demonstrate the PDC strategy and to validate the feasibility and effectiveness of the RSCS domain model. The results show that the restructured RSCS based on the proposed strategy and model can remedy the huge losses caused by the unavailability of raw materials.

Efficacy and safety of JAK inhibitor INC424 in patients with primary and post-polycythemia vera or post-essential

A phase II study (A2202) was performed to evaluate the efficacy and safety of JAK inhibitor ruxolitinib in 63 Chinese MF patients. Ruxolitinib was given twice a day (bid) at a starting dose of 15 mg (n=25) or 20 mg (n=38) based on a baseline platelet count. About 94.7% of the patients achieved a reduction in spleen size, 27.0% of which exhibited significant reduction (≥35%) at week 24. Significant improvement in debilitating constitutional symptoms, as assessed by MFSAF v2.0, was observed in patients treated with ruxolitinib. Ruxolitinib treatment was generally well tolerated by Chinese patients. Although the treatment was associated with an increase in certain adverse events (AEs) that were established as identified risks (anemia and thrombocytopenia), these AEs were considered manageable in this clinical setting. Ruxolitinib provided substantial reductions in splenomegaly and improvements in symptoms, and was well-tolerated by Chinese patients with MF.

Evaluation of endocrine disruption and dioxin-like effects of organic extracts from sewage sludge in

摘要: Study on effective disposal and utilization of sewage sludge has recently been the target of growing interest in China. However, potential risks are associated with the use of sludge due to its contamination with toxic organics, heavy metals and pathogenic microorganisms. In this study, a screening assessment was conducted on sewage sludge samples collected from 17 different sewage treatment plants in Beijing, based on a batch of in vitro bioassays, including a set of recombinant gene yeast assays for endocrine disruption, and an ethoxy resorufin-O-deethylase (EROD) assay using H4IIE cells for aryl hydrocarbon receptor (Ah-R) agonistic activities. Our results suggested that moderate levels of estrogen receptor agonistic activities (0.9 ng E2·g to 6.8 ng E2·g , dw), but relative higher androgen receptor antagonistic activities (nd to 45%), progestin receptor antagonistic activities (nd to 80%) and Ah-R agonistic activities (1390 to 6740 pg 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)·g , dw) were found in sewage sludge samples. However, there were no significant correlations between the toxic effects of sewage sludge and the sewage treatment processes. In addition, the 17β-estradiol (E2) equivalent quantity (EEQ) level of the sewage sludge was increased after the composting treatment, whereas the 2,3,7,8-tetrachlorodibenzo- -dioxin toxic equivalent quantity (TEQ) level of sewage sludge composted was much lower than that of sewage sludge.

Dynamic modelling and simulation of a post-combustion CO capture process for coal-fired power plants

摘要: Solvent-based post-combustion capture technologies have great potential for CO2 mitigation in traditional coal-fired power plants. Modelling and simulation provide a low-cost opportunity to evaluate performances and guide flexible operation. Composed by a series of partial differential equations, first-principle post-combustion capture models are computationally expensive, which limits their use in real time process simulation and control. In this study, we propose a first-principle approach to develop the basic structure of a reduced-order model and then the dominant factor is used to fit properties and simplify the chemical and physical process, based on which a universal and hybrid post-combustion capture model is established. Model output at steady state and trend at dynamic state are validated using experimental data obtained from the literature. Then, impacts of liquid-to-gas ratio, reboiler power, desorber pressure, tower height and their combination on the absorption and desorption effects are analyzed. Results indicate that tower height should be designed in conjunction with the flue gas flow, and the gas-liquid ratio can be optimized to reduce the reboiler power under a certain capture target.

Adiponectin: mechanisms and new therapeutic approaches for restoring diabetic heart sensitivity to ischemic post-conditioning

Systemic inflammatory response following myocardial ischemia-reperfusion injury (IRI) to a specific organ may cause injuries. Ischemic post-conditioning (IPostC) has emerged as a promising method for myocardial protection against IRI both in experimental and in clinical settings. Enhancement of endogenous nitric oxide (NO) is one of the major mechanisms by which IPostC confers cardioprotection. However, the sensitivity of the diabetic heart to IPostC is impaired and the underlying mechanism is unknown. Adiponectin (APN) is an adipocyte-derived plasma protein with anti-diabetic and anti-inflammatory properties. Plasma levels of APN are decreased in obese subjects and in patients with type 2 diabetes. APN supplementation has been shown to increase NO production and attenuate myocardial IRI in normal (non-diabetic) animals. However, the effect of APN on myocardial injury in diabetic subjects, especially its potential in restoring the sensitivity of the diabetic heart to IPostC has not been investigated. In the current paper, we discussed the possible reasons why the myocardium of diabetic subjects loses sensitivity to IPostC and also highlighted the potential effectiveness and mechanism of APN in restoring IPostC cardioprotection in diabetes. This review proposes to conduct studies that may facilitate the development of novel and optimal therapies to enhance cardioprotection in patients with severe diseases such as diabetes.

Enhancement of the polynomial functions response surface model for real-time analyzing ozone sensitivity

摘要: Abstract • The calculation process and algorithm of response surface model (RSM) were enhanced. • The prediction errors of RSM in the margin and transition areas were greatly reduced. • The enhanced RSM was able to analyze O3-NOx-VOC sensitivity in real-time. • The O3 formations were mainly sensitive to VOC, for the two case study regions. Quantification of the nonlinearities between ambient ozone (O3) and the emissions of nitrogen oxides (NOx) and volatile organic compound (VOC) is a prerequisite for an effective O3 control strategy. An Enhanced polynomial functions Response Surface Model (Epf-RSM) with the capability to analyze O3-NOx-VOC sensitivities in real time was developed by integrating the hill-climbing adaptive method into the optimized Extended Response Surface Model (ERSM) system. The Epf-RSM could single out the best suited polynomial function for each grid cell to quantify the responses of O3 concentrations to precursor emission changes. Several comparisons between Epf-RSM and pf-ERSM (polynomial functions based ERSM) were performed using out-of-sample validation, together with comparisons of the spatial distribution and the Empirical Kinetic Modeling Approach diagrams. The comparison results showed that Epf-RSM effectively addressed the drawbacks of pf-ERSM with respect to over-fitting in the margin areas and high biases in the transition areas. The O3 concentrations predicted by Epf-RSM agreed well with Community Multi-scale Air Quality simulation results. The case study results in the Pearl River Delta and the north-western area of the Shandong province indicated that the O3 formations in the central areas of both the regions were more sensitive to anthropogenic VOC in January, April, and October, while more NOx-sensitive in July.

摘要: -Glucuronidase from Li-3 (PGUS) can efficiently hydrolyze glycyrrhizin into the more valuable glycyrrhetic acid monoglucuronide. However, a low productivity of PGUS and the lack of an effective separation strategy have significantly limited its industrial applications. Therefore, the production of PGUS has been improved by optimizing both the fermentation and purification strategies. A two-stage fermentation strategy was developed where PGUS was first grown with glucose and then PGUS was produced in the presence of glycyrrhizin as an inducer. By using this strategy, the biomass was increased 1.5 times and the PGUS activity increased 5.4 times compared to that when glycyrrhizin was used as the sole carbon source. The amount of PGUS produced was increased another 16.6% when the fermentation was expanded to a 15-L fermenter. An effective protocol was also established to purify the PGUS using a sequential combination of hydrophobic, strong anion-exchange and gel filtration chromatography. This protocol had a recovery yield of 6% and gave PGUS that was 39 times purer than the crude PGUS. The purified PGUS had a specific activity of 350 U·mg .

Internet of Things (IoT)–blockchain-enabled pharmaceutical supply chain resilience in the post-pandemic

摘要: During the COVID-19 pandemic, the current operating environment of pharmaceutical supply chain (PSC) has rapidly changed and faced increasing risks of disruption. The Internet of Things (IoT) and blockchain not only help enhance the efficiency of PSC operations in the information technology domain but also address complex related issues and improve the visibility, flexibility, and transparency of these operations. Although IoT and blockchain have been widely examined in the areas of supply chain and logistics management, further work on PSC is expected by the public to enhance its resilience. To respond to this call, this paper combines a literature review with semi-structured interviews to investigate the characteristics of PSC, the key aspects affecting PSC, and the challenges faced by PSC in the post-pandemic era. An IoT–blockchain-integrated hospital-side oriented PSC management model is also developed. This paper highlights how IoT and blockchain technology can enhance supply chain resilience and provides a reference on how PSC members can cope with the associated risks.

CRISPR-Cas9 mediated LAG-3 disruption in CAR-T cells

T cells engineered with chimeric antigen receptor (CAR) have been successfully applied to treat advanced refractory B cell malignancy. However, many challenges remain in extending its application toward the treatment of solid tumors. The immunosuppressive nature of tumor microenvironment is considered one of the key factors limiting CAR-T efficacy. One negative regulator of T cell activity is lymphocyte activation gene-3 (LAG-3). We successfully generated LAG-3 knockout T and CAR-T cells with high efficiency using CRISPR-Cas9 mediated gene editing and found that the viability and immune phenotype were not dramatically changed during in vitro culture. LAG-3 knockout CAR-T cells displayed robust antigen-specific antitumor activity in cell culture and in murine xenograft model, which is comparable to standard CAR-T cells. Our study demonstrates an efficient approach to silence immune checkpoint in CAR-T cells via gene editing.

面向2035 年的灾害事故智慧应急科技发展战略研究

摘要:

智慧应急科技发展是公共安全保障的重要组成部分,也是实现国家治理现代化、建设更高水平的平安中国的应有之义。本文研究旨在深化 2035 年远景目标,进而为我国灾害事故智慧应急发展的系统规划和前瞻部署提供支撑。本文以灾害事故智慧应急科技发展需求为导向,全面分析了国际态势和主要发展趋势;重点从自然灾害防御、事故灾难防范、应急平台建设等方面着手,对我国相关领域发展的基本状况和面临的问题进行剖析。在此基础上,紧密结合新一代信息技术的发展,提出了我国灾害事故智慧应急科技发展的总体思路框架与战略方向,涵盖重要科技发展方向、重点基础研究方向、重大科技研发任务等;针对我国自然灾害和事故灾难的智慧应急体系与能力现代化建设的科技发展路径,从政策和理论体系研究方面提出了相应发展建议。

Parametric study of hexagonal castellated beams in post-tensioned self-centering steel connections

摘要: The effects of important parameters (beam reinforcing plates, initial post-tensioning, and material properties of steel angles) on the behavior of hexagonal castellated beams in post-tensioned self-centering (PTSC) connections undergone cyclic loading up to 4% lateral drift have been investigated by finite element (FE) analysis using ABAQUS. The PTSC connection is comprised of bolted top and bottom angles as energy dissipaters and steel strands to provide self-centering capacity. The FE analysis has also been validated against the experimental test. The new formulations derived from analytical method has been proposed to predict bending moment of PTSC connections. The web-post buckling in hexagonal castellated beams has been identified as the dominant failure mode when excessive initial post-tensioning force is applied to reach greater bending moment resistance, so it is required to limit the highest initial post-tensioning force to prevent this failure. Furthermore, properties of steel material has been simulated using bilinear elastoplastic modeling with 1.5% strain-hardening which has perfectly matched with the real material of steel angles. It is recommended to avoid using steel angles with high yielding strength since they lead to the yielding of bolt shank. The necessity of reinforcing plates to prevent beam flange from local buckling has been reaffirmed.
